Le ms. a appartenu à Claude Dupuy (1545-1594), puis à ses fils Pierre et Jacques Dupuy. Claude Dupuy a laissé plusieurs notes dans le ms. (1: table, 101v-105: numéros des odes d'Horace glosées en marge, 139v: mention de lacune, biffée après acquisition du cahier manquant), et Pierre a inscrit le titre au dos de la reliure de parchemin. Le 26 mai 1652, après la mort de Pierre, Jacques lègua toute leur bibliothèque par testament à la Bibliothèque du roi. Ces volumes y entreront en 1657 après sa mort le 17 novembre 1656. Les 260 manuscrits anciens font partie du legs (voir Delisle I, p. 335-336 et Omont, Concordances, p. 77-78).
